FIMM Genomics Sanger sequencing sample delivery

Here are short instructions on sample preparation and delivery for samples coming in for Sanger sequencing or fragment analysis. More detailed instructions for sample prep are available upon request via email.
Full service Sanger sequencing samples
  • Customer provides 6,6ul of template – primer mix on V-bottomed PCR plates or PCR strips (use strips only for small number of samples, do not use individual tubes) and a fully filled in sample form submitted via iLab.
  • Samples should be pipetted on the plate or strip in columns starting from well A1 -> H1, then A2 -> H2 and so on, and listed on the sample form in the same exact order. Sample names should not contain any special characters. Please see the sample form template for full instructions.
  • Label your plates with customer name and service ID. Label your strips with orientation, for example mark wells A1 A2 A3… on the top of each strip or use a running number to differentiate between strips. Use the sample information stickers on the sample fridge door and place a sticker on top of your samples. SeqLab provided plate racks can be found in a basket on the table next to the sample fridge – feel free to use these to secure your strips in the fridge.
  • Plates or strips must be sealed carefully to avoid sample evaporation. Use plate seals with strong adhesive or PCR strip caps. Do not use individual strip caps or heat-seal!
  • Spin down plates or strips before delivery (see instructions below).
  • Turnover time for full service samples is 2-3 business days for full plates and up to a week for samples sets smaller than a full plate.
Ready to run Sanger sequencing samples
  • Customer provides 10ul of sequencing reactions on V-bottomed PCR plates or PCR strips (use strips only for small number of samples, do not use individual tubes) and a fully filled in sample form submitted via iLab. If sample has evaporated significantly during sequencing reaction, please top up with water to vol 10ul.
  • Spin down plates or strips before starting the sequencing reactions as air bubbles at the bottom may interfere with the reaction.
  • Samples should be pipetted on the plate or strip in columns starting from well A1 -> H1, then A2 -> H2 and so on, and listed on the sample form in the same exact order. Sample names should not contain any special characters. Please see the sample form template for full instructions.
  • Label your plates with customer name and service ID. Fragment Analysis plates

Fragment analysis run is performed on full 96-plates – each well must contain at least 10ul of HiDi Formamide and size standard mix.

  • Samples must be delivered on Applied Biosystems MicroAmp™ Optical 96-Well Reaction Plate with Barcode (catalogue number 4306737).
  • Plates must be sealed carefully and protected from light. Spin down plates before delivery (see instructions below).
  • Turnover time for fragment analysis plates is typically 1-2 business days.
General sample delivery instructions
  • All samples coming in for Sanger sequencing or fragment analysis can be delivered at room temperature. SeqLab sample fridge is located in Biomedicum 2U, 2nd floor, F-wing, room F216.
  • You can bring your samples directly to the SeqLab sample fridge, drop them off at the Biomedicum 2U info desk, or send them by mail/courier.
  • If you bring your samples to our sample fridge, please use the sample stickers on the fridge door to label your samples. Use the same customer name and Service ID as you have used on your service request in iLab. Place the sticker on top of your sample set.

  • Please note that it may take up to two weeks for your samples to arrive in our lab if they are sent by regular mail (Posti) - if you need your results urgently do consider using a courier service. 
  • If you send your samples using the UH internal mail service, please write SAMPLES or NÄYTTEITÄ on top of the letter so that it will be forwarded to the info desk.
