When your iLab service request has been approved, you can contact core staff and schedule sample delivery.
General RNA sample requirements

RNA samples

  • Samples should be delivered in a skirted 96-well plate in column wise orientation (A1, B1, C1...) marked with a date and the project name (sample batch ID in iLab) 
    • There should be no empty wells or columns between samples. Full reaction cost will be charged for empty wells.
    • Recommended plate: skirted Eppendorf™ twin.tec™ 96 Well LoBind PCR Plate (Catalog no. 0030129512)
    • Plates must be sealed carefully with a PCR plate seal 
    • Sample delivery on dry ice
    • Sample information including sample ID, well, concentration in ng/ul, volume, source material, and possible batch information through iLab
    • Reference genome information
  • We have no capacity for long time sample storage so delivered RNA samples are for one-time use only.
Application specific sample requirements

Illumina Stranded Total RNA Prep with Ribo-Zero Plus

  • DNase I -treated total RNA samples in at least 20 µl of nuclease free-water
    • Minimum concentration of 5 ng/µl of good quality (RIN ≥ 8, Agilent Bioanalyzer) total RNA
    • DNA contamination of the samples should be less than 10%
    • Library prep input amount should be 10 - 1000 ng 

Illumina Stranded mRNA Prep 

  • DNase I -treated total RNA samples in at least 30 µl of nuclease free-water 
    • Minimum concentration of 5 ng/µl of good quality (RIN ≥ 8, Agilent Bioanalyzer) total RNA
    • DNA contamination of the samples should be less than 10%
    • Library prep input amount should be 50 - 1000 ng (recommended input 500 ng)
    • The kit does not accept purified mRNA as input

Quantabio sparQ RNA-Seq HMR kit

  • Total RNA samples in 20 µl nuclease-free water
    • Minimum concentration of 0.2 ng/µl of good quality (RIN ≥ 7, Agilent Bioanalyzer) total RNA. For degraded and/or FFPE RNA 2 ng/µl of total RNA.
    • DNA contamination of the samples should be less than 10%
    • Library prep input amount should be 1 - 1000 ng for high-quality total RNA and 10-1000 ng for degraded and/or FFPE RNA 

Watchmaker RNA Library Prep Kit with Polaris™ Depletion

  • Total RNA samples in 30 µl nuclease-free water
    • Minimum concentration of 0.1 ng/µl of good quality (RIN ≥ 7, Agilent Bioanalyzer) total RNA. For degraded and/or FFPE RNA 1 ng/µl of total RNA.
    • DNA contamination of the samples should be less than 10%
    • Library prep input amount 1 - 1000 ng for high-quality total RNA or 10 – 1000 ng for partially degraded and/or FFPE RNA

Lexogen Small RNA-seq library prep

  • DNase I -treated total RNA samples with miRNA fraction in 20 µl of nuclease-free water
    • Minimum concentration of 5 ng/µl of good quality (RIN ≥ 8, Agilent Bioanalyzer) total RNA
    • DNA contamination of the samples should be less than 10%
    • Library prep input amount should be 50 pg – 1000 ng (recommended input ≥100ng)

SMART-Seq mRNA 

  • Total RNA samples in 20 µl nuclease-free water
    • Minimum concentration of 0.1 ng/µl of good quality (RIN ≥ 8, Agilent Bioanalyzer) total RNA
    • DNA contamination of the samples should be less than 10%
    • Library prep input amount should be 10 pg – 100 ng 

PacBio Kinnex full-length RNA kit

  • Total RNA samples in 20 µl nuclease-free water
    • Minimum concentration of 50 ng/µl of good quality (RIN ≥ 7, Agilent Bioanalyzer) total RNA. 
    • Library prep input amount 300 ng

Oxford Nanopore cDNA-PCR Sequencing V14 library prep

  • Total RNA samples in 20 µl nuclease-free water
    • Minimum concentration of 50 ng/µl of good quality (RIN ≥ 9, Agilent Bioanalyzer) total RNA. 
    • Library prep input amount 500 ng
