Leica TCS SP8 STED 3X CW 3D is an inverted confocal microscope equipped with motorized stage. It can be used to acquire 3D images from cell or tissue samples or individual images from given focal plane without distortions from unfocused signal. You can also aqcuire cross section images. Motorized stage can be used to acquire timelapse images from multiple locations as the system's in-built autofocus ensures that your sample stays in focus for the duration of your experiment. The microscope is equipped for live imaging.
The system can be used to aqcuire super-resolution images using the 592 nm depletion laser using the 93x glycerol objective. One color STED is possible with almost any green color, but two color imaging requires very specific dyes to work.
Room 2036a
SP8 Upright is a fully motorized upright confocal. It has four laser lines and three decetors as well as motorized stage with Leica Navigator. This allows the microscope to be used to reconstruct 3D structures of both cells and tissue, though due to the number of detectors imaging 3D samples with more than three colors tends to be slow. The system does not have enviromental control and thus cannot be used with live samples that require heating or CO2.
Notably the 25X 0.95 NA dipping water objective is excellent for large plant tissue that is imaged in a dish. However the system lacks the high-magnification water objective that is often used to study Arabidopsis.
Room 2036b
Leica Stellaris 8 is a next-generation confocal microscope with white light laser and high-end detectors. The system can record lifetimes on the go, which allows the elimination of the background signal as long as the background and the actual signal have sufficiently different lifetimes. The in-built resonant scanner allows fast imaging with relatively low photo-toxicity when needed. Stellaris 8 also has a motorised stage which allows you to combine images together with tile imaging to take an image of a large area. The microscope is also equipped for live imaging.
Stellaris 8 be used to perform FLIM experiments as well as measure FRET signal in the sample if sufficient controls have been prepared.
Room 2027
Olympus FV4000 is an inverted confocal microscope equipped with a motorized stage. It can be used to acquire 3D images from cell or tissue samples or individual images from a given focal plane without distortions from unfocused signal. The motorized stage can be used to acquire timelapse images from multiple locations. FV4000 has silicon oil objectives, making it particularly good for tissue samples. Stabilized lasers and photon-counting detectors allow the user to quantify the signal intensity with high fidelity.
The microscope is equipped for live imaging.
