Custom Vector Cloning

We offer custom cloning services using Gibson assembly, enabling scarless and flexible construction of DNA plasmids. This method allows us to assemble multiple DNA fragments without relying on restriction enzyme sites.

We support cloning into AAV‑compatible vectors as well as other custom vector backbones.

Cloning into AAV Expression Vectors

Through our collaboration with the unit, we can clone your cDNA into AAV‑compatible expression vectors and support downstream AAV production.

You may:

  • Choose a cDNA from , or
  • Provide your own plasmid

Available promoters: CAG or CMV.

Service includes:

  • Gibson assembly cloning into your selected AAV vector
  • Verification of the insert by Sanger sequencing
  • Preparation of a DNA midiprep of the final construct
  • Transfer of the cloned plasmid to HelVi‑AAV for virus production

Cloning into Custom Vectors

Gibson assembly allows the integration of multiple fragments into virtually any vector backbone.
This enables a wide range of custom cloning applications, such as:

  • Adding one or more tags to your cDNA
  • Introducing a specialized or alternative promoter
  • Making a vector Gateway‑compatible
  • Replacing or modifying existing regulatory elements
  • Other custom design requests
