3I Marianas (3I intelligent Imaging Innovations) is a fluorescence microscope designed for imaging live cells with multiple colours and techiques. Available imaging techniques are wide-field, TIRF and spinning disk confocal. There are nine light sources (5 LEDs and 4 lasers) and fast switching times between imaging techniques and light sources so that one can make almost any combination of videos. Additionally it is possible to do photomanipulation experiments (FRAP an photo activation) . Microscope has a motorised stage which can be utilized for multiposition imaging or image plates.

Cell-IQ is a continuous cell culturing platform for cell lines, primary cells, co-cultures and monolayer tissue models. It identifies, visualizes and records in real time all changes in morphology and physiology down to the single cell during an incubation and translates the images into graphical outputs. We have two units. One with phase contrast and 3 channel fluorescence immaging and the other with only phase contrast imaging.

Leica DM6000B is an upright fluorescence wide field microscope that can also aqcuire bright field, polarisation and DIC images. It is meant for fixed samples labeled with fluorescence or histological labels. Polarisation and DIC contrast can be used to acquire images from samples without labels. Motorized stage makes it possible to image big areas with good resolution while 1,25x and 2,5x objectives can be used to aqcuire an image field quickly in one go when using transmitted light. Stage has capacity either for four slides or for one 127x85mm plate.

It can be used to acquire 3D images from cell or tissue samples or individual images from given focal plane without distortions from unfocused signal. You can also aqcuire cross section images. Microscope is not equipped with incubation chamber.

SP5II HCS is a fully motorized inverted point scanning confocal. It is especially good for high resolution imaging for live samples without blur from out of focus signal. Scanner has two extra sensitive low noice detectors for dim samples and faster imaging.

Microscope has a motorised stage for multipoint or tile imaging and the system has a Leica Matrix software for high content imaging from plates. You can also aqcuire cross section images.

Multiphoton

Leica TCS SP5 MP, an upright (objectives are above the sample) confocal microscope with tunable multiphoton laser.  It can be used to acquire 3D images from cell or tissue samples or individual images from given focal plane without distortions from unfocused signal. You can also aqcuire cross section images from your samples. 

Multiphoton laser can be used to reach deep into tissue samples or alternatively to generate second harmonic images from suitable structures. Microscope is also equipped for live imaging.

FLIM and Fluorescence (cross-)correlation spectroscopy

Microscope is equipped with sensitive SPAD detectors and photon counting system allowing fluoresence lifetime (FLIM) imaging as well as fluoresence cross correlation spectroscopy (FCS). These techniques are primarly used to study protein interactions in cells.

Leica TCS SP8 STED 3X CW 3D is an inverted confocal microscope equipped with motorized stage.  It can be used to acquire 3D images from cell or tissue samples or individual images from given focal plane without distortions from unfocused signal. You can also aqcuire cross section images.  Motorized stage can be used to acquire timelapse images from multiple locations as the system's in-built autofocus ensures that your sample stays in focus for the duration of your experiment. The microscope is equipped for live imaging. 

STED

The system can be used to aqcuire super-resolution images using the 592 nm depletion laser using the 93x glycerol objective.  One color STED is possible with almost any green color, but two color imaging requires very specific dyes to work.

Cellomics CellInsight is a compact plate reader. It accepts most well-plate types and there is an option to  design custom sample geometries for e.g. micro arrays. Due to the lack of CO2 chamber and heating the system does not suit live cell work.

For image analysis, we recommend CellProfiler. We do have a license for Thermo Bio-Applications for running image analysis either during scanning, or later on (disk-based rescan), but this software is only available on the instrument PC.

Our CellInsight is not working perfectly. There are random imaging errors where channels are either switched or missing altogether. According to Thermo, spare parts are no longer available for this instrument. You can find a more reliable CellInsight at BIU.

Lightsheet imaging is a fluorescence microscopy technique with an intermediate optical resolution but good optical sectioning capabilities and high speed.  This means that the device is best employed to study large structures such as organs and embryos.  It is also especially good for sensitive live samples because the intensity of the light is low comparing to other fluorescence imaging technics. Method is only suitable for fluorescence imaging.

LSM 700 is a basic upright laser scanning confocal microscope. It can be used to acquire 3D images from cell or tissue samples or individual images from given focal plane without distortions from unfocused signal. Microscope has a motorised stage which allows you to combine images together with tile imaging to take an image of a large area.  The microscope is equipped for live imaging. The system has no transmitted light detector so bright field images cannot be aqcuired.

LS workstation is a powerful workstation for 3D image analysis and deconvolution (Microvolution) for large data sets. It has a fast network connection (10Gbit) for big data transfer from network disk. This workstation has a limited Imaris software package. ImarisXT module allows you to use Matlab and Python scripts and Fiji modules within Imaris.

Room 2011b

HCA Workstation is an older workstation with fast disk and some offline microscope software. It is suitabler for usage where you don't need 3D visualisation, high computing power of lot of local disk space. Its major usage is analysing high content data from CellInsight and Fluorescence lifetime imaging data from SP5MP FLIM.
HCA Workstation has a variety of software for image analysis including dedicated tools for analysing high content imaging projects.
