Centre for Virus and Macromolecular Complex Production (ICVIR)

Instruct is one of the successful operators at the European Strategy Forum on Research Infrastructures (ESFRI) for integrated structural biology infrastructure for Europe where major distributed research infrastructures supported by member states offer European wide access to scientifically evaluated and accepted Instruct Centres across Europe. Instruct Ltd. is a landmark European Research Infrastructure on the ESFRI Roadmap (2016). Instruct Ltd. has entered the European Research Infrastructure Consortium (ERIC) on the 4th July 2017 to form Instruct-ERIC that will gradually replace the current Instruct Ltd.

Instruct-ERIC provides peer-reviewed access to all of the core structural biology technologies for excellent scientific projects, developing the next generation of instrumentation in partnership with industry, and providing training for structural biologists in cutting edge techniques.

Instruct-FI is a national, integrated, structural cell biology network that supports state-of-the-art research in protein, virus and macromolecular complex production, purification and characterization; high resolution cryo-electron microscopy (cryoEM), nuclear magnetic resonance (NMR), native mass spectrometry (MS), structural bioinformatics and X-ray crystallography (X-ray). Instruct-FI activities are distributed in six universities throughout Finland (University of Helsinki, University of Oulu, University of Turku, Åbo Academi, University of Eastern Finland, University of Tampere). All units are well-established as scientifically-driven core facilities with wide industrial and academic, international and domestic user networks. Instruct-FI is included on the Finnish Research Infrastructure 2014-2020 roadmap. The Academy of Finland Research Infrastructure Committee has recommended (31.1.2017) that Finland applies full membership in Instruct-ERIC. Instruct-FI will apply as a new distributed Instruct-ERIC Centre. Currently in Instruct Ltd., Instruct-FI has a national affiliate centre status and one of its nodes, Instruct Centre for Virus and Macromolecular Complex Production (ICVIR), is an approved Instruct Ltd Centre.

Instruct-FI aims to organize open access services for the community. Instruct-FI provides access to equipment and expertise at two levels: (i) guiding new users through unfamiliar structural cell biology technologies that can extend their research and open new opportunities and (ii) facilitating expert users to access specialist, cutting-edge technologies.

Our Instruct Centre for Virus and Macromolecular Complex Production (ICVIR) operates in the field of large scale virus and macromolecular complex production and development of purification methods for their structural analysis. The unit provides purification technologies and expertise specializing in macromolecular complex sample preparation for functional and structural studies (BSL 1-2). The centrifugation platform (rate-zonal, equilibrium, flotation and differential centrifugation) is equipped with a farm of ultracentrifuges, superspeed centrifuges and the corresponding swinging out and fixed angle rotors with different capacities (Sorvall and Beckman) as well as gradient making and fractionation instrumentation. For chromatographic purification of macromolecular complexes, monolithic technology (BiaSeparations) and asymmetric flow field flow fractionation (AF4, Postnova Analytics) service technologies are provided. All techniques can be used in different combinations to purify macromolecular complexes, such as viruses, virus-like particles, vesicles etc. Senior staff guides the users in sample preparation.
